It's the timing more than anything that has him on his guard: the very sudden offer to trim off some rations-fat directly after the announcement of the death of his partner. He's well-aware of the fact that Kyouya deciding to eat means very little when Kyouya had been the one to set the bowls down, and even though it may just be excessive paranoia, he's survived enough attempts on his life throughout the years to trust his gut instinct.
He could state his suspicions on the spot and more openly refuse the (admittedly fairly generous, possibility of foul-play aside) gifted ration. That isn't how Jack tends to play the game, though. He smiles, freeing up one of his hands to brush hair out of his eyes.]
